What is OTOSPORIN

OTOSPORIN is a combination ear drop medication used to treat external ear infections (otitis externa). It contains three active ingredients: Neomycin sulphate (an aminoglycoside antibiotic), Polymyxin B sulphate (a polypeptide antibiotic), and Hydrocortisone (a corticosteroid). This formulation is effective in treating infections caused by susceptible bacteria while simultaneously reducing inflammation, pain, and itching.

• Neomycin sulphate: Bactericidal action against gram-positive and gram-negative organisms.
• Polymyxin B sulphate: Potent activity against gram-negative organisms including Pseudomonas aeruginosa.
• Hydrocortisone: Reduces inflammation, redness, and swelling in the ear canal.

How to use OTOSPORIN

Use OTOSPORIN exactly as prescribed by your healthcare provider. It is intended for otic (ear) use only.

• Clean and dry the affected ear before applying the drops.
• Warm the bottle by holding it in your hand for a few minutes to reduce the sensation of coldness in the ear.
• Lie down or tilt your head to one side with the affected ear facing upward.
• Instill the prescribed number of drops (usually 3–4 drops) into the ear canal.
• Keep your head tilted for several minutes to allow the medicine to reach deep into the canal.
• Avoid touching the dropper tip to prevent contamination.
• Use it typically 2–4 times daily as prescribed.

Mode of Action OTOSPORIN

Each component of OTOSPORIN contributes to the overall therapeutic effect:

• Neomycin: Inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, leading to bacterial cell death.
• Polymyxin B: Interacts with bacterial cell membranes, altering permeability and causing leakage of cellular contents.
• Hydrocortisone: Binds to glucocorticoid receptors, reducing inflammatory mediators like prostaglandins and leukotrienes, thereby reducing swelling, redness, and itching.

The synergistic combination ensures comprehensive antimicrobial coverage while alleviating symptoms of inflammation.

OTOSPORIN Interactions OTOSPORIN

OTOSPORIN is topically applied, so systemic drug interactions are minimal. However, caution is advised in the following:

• Concurrent use with other ototoxic drugs (e.g., loop diuretics, aminoglycosides like gentamicin) may increase the risk of hearing damage, especially in patients with tympanic membrane perforation.
• Avoid combining with other topical ear preparations unless advised by a healthcare provider.
• Do not use concurrently with systemic corticosteroids unless directed, as it may potentiate systemic effects.

Always inform your healthcare provider about any medications you are taking, including over-the-counter drugs and supplements.

Dosage of OTOSPORIN

The dosage must be individualized based on the severity of infection and medical history.

• Typical adult dose: 3–4 drops in the affected ear 2–4 times a day.
• Children: Dosing is similar, but always follow pediatrician instructions.
• Course of treatment usually lasts 7–10 days.

Do not exceed the prescribed dosage. Prolonged use can increase the risk of fungal or secondary bacterial infections.

Possible side effects of OTOSPORIN

Most patients tolerate OTOSPORIN well, but side effects may occur in some cases.

• Common side effects:
  • Local irritation or stinging in the ear
  • Itching or redness of the ear canal
• Serious side effects (rare):
  • Allergic reactions (rash, swelling, difficulty breathing)
  • Ototoxicity (hearing loss, tinnitus) especially with prolonged use or tympanic membrane damage
  • Secondary fungal infections due to long-term corticosteroid use

If any unusual symptoms occur, discontinue use and consult a healthcare provider immediately.

OTOSPORIN Contraindications OTOSPORIN

OTOSPORIN should not be used in the following conditions:

• Known hypersensitivity to neomycin, polymyxin B, hydrocortisone, or any component of the formulation
• Presence of perforated tympanic membrane (risk of ototoxicity)
• Viral infections of the ear canal, such as herpes simplex or varicella
• Untreated fungal or tuberculous ear infections

Careful assessment is necessary before prescribing to individuals with a history of ear surgery or chronic ear conditions.

Storage of OTOSPORIN

Proper storage ensures the medicine remains effective and safe for use.

• Store at a temperature below 25°C (77°F).
• Keep the bottle tightly closed when not in use.
• Do not freeze.
• Protect from direct sunlight and moisture.
• Keep out of reach of children.
• Discard any unused solution after 28 days of opening.

Check the expiry date before use and do not use beyond it.
